Going to join the large tank club, but have planning questions

Buying a new house, and with a new house, the best part is the new tank. I have already planned a lot of the details, for example I am going to go with a plywood tank. I also already have the majority of the equipment purchased (all I need is sand, and one more pump).

So here is what I am planning:
It will be an in wall system, committing most of a room behind it to house the tank stuff.

I am expecting it to be about 375 to 450gal in size. The largest being 72x48x32

So on to the first questions:

1) how can I tell if a wall is load barring?

2) I assume I can't put a tank this large on top of carpet? The room I am right now thinking of placing the tank in, is currently carpeted. I plan to be in the house for a long time, but I hate to cut the carpet out of a room if I don't have to. I want to make design things in a way to not reduce resell (terribly) in the future. Any thoughts on how to do this best?

I am sure I will have more questions in the future, I appreciate any input.
