Head height question

So I'm going to have my sump in a separate room than my tank. Should I calculate the length of my PVC going horizontally to the tank when I count my head height.

You need the height from water surface to water surface for the lift portion of losses. Knowing to total length of the pipe is for knowing the friction losses. If you know the flow rate there are lots of charts you can look up to see about what you have. You can estimate the losses per fitting by adding 1ft of pipe run per fitting, not 1ft of losses per fitting.
