I assume bulkheads are going in these holes, and if so the distance is limited by the flange. But even if not for bulkheads you can get them pretty close. As you can see I have drilled 4" hole pretty close to one another. Just take your time.

no. you should leave a minimum of one hole diameter between edges (or 2 diamteters center to center,) preferably 1.5x the diameter edge to edge. The same applies from the edge of the tank glass to the edge of a hole.
