2 months should about do it - although 72 days fallow would be optimal.
If you are going to treat with Cupramine, you don't want to use any rock or substrate as those will absorb some of the copper and make it difficult to maintain a stable level. You will have to control the ammonia either by doing WCs, or if the HOB utilizes bio-media which can "host" bacteria then you can use something like Bio-spira to "seed" it.
