The skimmer will only remove copper in proportion to the amount of liquid collected - so it is minimal. If you remove a cup of skimmate from a 50 gallon tank, you'll remove 1/800 of the copper dose. I've routinely used skimmers on FO tanks with copper and never had any issues.

Cupramine has such a wide range between 'effective" and "deadly to fish", that you have a little room for error with the testing. But, because of the LR, you never really know how much copper is in the water column, so frequent testing is needed. I really prefer the SeaChem test kit; its made by the same mfg as the Cupramine and that seems to make sense to me. Cupramine won't kill much of your bio-filter, but every invert in the tank will go, so keep an eye on ammonia. Although I don't think you'll have a problem in a large tank. Since its a FOWLR I decided to do treatment on the display. I know the live rocks would absorb a lot of Cupramine but Im not sure about the skimmer. I leave it off for an hour when adjusting dose then turn back on again.
Im using both Salifert and Seachem for testing copper level. I take the 20 minutes reading on the Salifert. Any advice is appreciated.
I have a 400 fowlr system that I treated with cupramine and it was a pain it took me a week to get a constant .5 and yes I did leave my skimmer off I had a slight ammonia spike but went away in a few days and the blue legs I forget to get out lived through the whole treatment.
