Probably not, but it may go through a mini cycle.
As you know, cycling is really about matching filtering bacteria population to the bio load. If you add a lot of livestock at once and overshoot the filtering capacity of your LR and LS, your system will have some ammonia to deal with. I don't think your tank will go through a full blown cycling with a high ammonia count as long as you have kept the rocks and sand in a good condition through the transfer.
I'd go slow on moving livestock into the new tank if you can. Test for ammonia and nitrate as you add more animals. Can you get some macroalgae to throw into your sump/refugium? They will help you through the initial period since they will consume ammonia and nitrate.
