i have just filled my 75 gallon tank with ro water the sand is in, I am about to mix the salt, I was wondering if I should have filters on or off while I mix the salt

Its best to mix the salt/water in buckets IMO also if you were going to mix the salt in the tank you shouldn't have put the sand in yet.
