when to test water

i just did a 20% water change and replaced with RO/DI water. when should i test. should i do it now or wait till morning?

i added salt and the salinity is fine. i need to check my nitrates they were 20 this morning. I lost both my clowns. i figure that was due to stress with me rearangeing the corals and rock.

depending on tank turn over 30min to a hour

Nitrates of 20 or rearranging your rock shouldn't kill clownfish.
