Artemia decapsulation.

Hi everyone,

Can anyone tell me how I can preserve decapsulated artemia eggs please?
Atm I decapsulate daily and would like to be able to decapsulate a larger amount, store, and use as and when required.
I have tried MgCl26H2O ( Magnesium Chloride) but that didn't seem to work.
Thanks, RB

Oh dear.

Nothing fancy. Just use salt. Keep the decapped cysts in a little water to which you add salt until no more will dissolve, and you can see the white crystals. Refrigerate. Simple.

I seen some info on decapsulateing Eggs yourself, I have some Premium Grade 90% from Brine Shrimp Dirtect, Are these decapsulated or should I do it for clown larva, if so any tips....Thanks Carl

From Hoff and Snell (5th Edition, 2001) ...

For long term storage, decapsulated cysts must be dehydrated. Dehydration of decapsulated cysts is accomplished by transferring 1 g of decapsulated cysts into 10 mL of saturated brine (330 g NaCl / L) and aerating for 18 hours. Due to the osmotic imbalance, cysts release water to the brine solution which should be replaced every 1-2 hour(s) or more salt added. After 18 hours, cysts have lost about 80% of their cellular water. Aeration can be stopped, the cysts settled and collected by filtration. The dehydrated, decapsulated cysts can now be transferred to a container, topped with fresh brine, sealed and stored in a refrigerator or freezer. Decapsulated cysts with 16%-20% cellular water content can be stored for a few months without decrease in hatching rate or percentage.
