I used two in my 180 and they worked great. In fact I still use the two for my nighttime flow in our 350G tank. Remarkable products.
I put them on opposite sides of the tank; one on the right side near the front, and the other on the left side near the rear. This results in a circular flow pattern that looks like a "race track" in the tank.

I'm in the process of setting up a 215 gal. and I'm going with two MP40s one on each end and one MP20 in the center of the back wall and I'm hoping that will be enough flow for my tank. So to answer your question; in my opinion you should use at least two MP40s.
