Super Glue while submerged

Noob question...

Can you use gel superglue IN water? I don't mean take a frag plug out of water, put glue on it, then put it back in...

There is a pre-drilled hole in my nano tank's overflow that I need to fill. It's about 3/4 of the way down and pulls quite a bit of water through it rather than the top overflow.

Do I need to drain the tank, dry the area off, glue, then fill?

I would rather stick some glue on it and be done. I thought about trying it with silicone, but curing silicone underwater in a small tank is a really bad idea. Superglue is by far the fastest and safest way.
 
Just put a nic3e bead of gel around the plug, insert it and be done. It might not seal perfectly. Weldon 16 would work better, but I'm not sure if it's reef safe before curing.
 
Sorry, should have been more descriptive.

Nano (5 gallon)
hole is about 1/4" x 1/2"

2 good dabs of superglue should do it.
