Will starting a refugium cause a small cycle? Isn't it like starting a small tank?

Im going to add one to a 150g aquarium. I was just taking caution.

No, it won't. There wouldn't be anything to trigger a cycle and even if there were, in all likelihood, the biological filtration that has been established in your reef with its live rock, skimmer, etc. would, unless there were something unforseeably wrong, be able to handle it. In other words...go ahead and don't worry.

How big is it going to be? And how much live sand are you going to add?

I have a 30g all-glass aquarium that I am going to install baffles in. I want to put mud then a layer of live sand. Would this benefit in any way. Or should I choose one or the other?

I'd chose either the mud or the live sand, but you don't really need either if you're going to have 1.5-pounds or more of live rock per gallon of the tank size, and a good quality skimmer. With that, you'd have all the "filtration" you'd need...particularly since your sump is going to include a refugium.
