what is a good way to get DNA from a live fish without cutting the fin? im guessing swabbing the mucus will yield some cells. thanks
Thats a very ambitious project. Unless you're already very good at cloning and know the DNA sequence of your gene of interest, then you're in over your head. It ain't gonna happen. I'm a PhD candidate in Cell Biology, and that would be an intimidating task for any PhD level research scientist, much less a high school student.
For starters, you don't really clone stuff from chromosomes. You'd need to isolate the messenger RNAs (b/c you can't have introns in your cloned gene), and then use those to generate a cDNA library. You'd need to know your sequence of interest in order to synthesize PCR primers, etc . . . it's exponentially more complicated than you'd think.
