If you use 2 mil bags, go with triple bagging, if 3 mil, double bagging should be good. Depending on what you are sending, it is good to float some frags (e.g. acros) on a little chunk of styro that is attached with a rubber band. Others are ok to set on the bottom of the bag. If they are long enough, can tie the bags. No need to leave air in the bag, and tie with just a little slack/flexibility in the bag. Ice packs are a must this time of year. Not sure if FedEx does home pick ups anymore, but used to have them pick up at the house or office as late as possible. Send priority overnight (guaranteed by 10:30 am).

Good thick styro box, lined with several layers of newspaper. If using bags, I triple bag frags and other things that might poke holes in them. For frags 1/3 water and 2/3 air will be fine. For small frags, those urine specimen cups that gatorlum69 suggested are excellent...tape the lids to be sure they don't unscrew. Also through in a couple of ice packs. This time of year with current temps, I'd use 4 ice packs in a standard fish shipping flat box. Wrap the ice packs in a sheet of newspaper, keeps them from making direct contact with any specimens and over chilling anything. Also pack as late as possible to bring to FedEx as late as you can and still make it in time for overnight delivery.

Right on the money, don't let the ice pack touch the bags, wrap the ice pack with newspaper, make sure that you use peanuts or other packing materials to ensure that the bags are snug, try if possible to use a good plastic liner inside or outside the styrofoam insert, also once you place the lid use the tape to seal the lid all around.
it wouldn't hurt to add stickers that indicate fragile and live animals. open an account with fedex or UPS and try to get a volume discount, Good luck.

Rogger hit a couple of things I forget.

Though with labeling things live animals or fish, only do that if you already have an account with the shipper that has been approved for shipping such. Otherwise you sometimes find your "live fish" shipment sitting back on your doorstep with a note saying they can't ship such things. That is how I found out about the need for prior approval of live animal/fish shipping. FedEx actually makes you ship their facility in Memphis a test package...bags of empty water, to see if you pack to their standards. Their standard being to avoid leaking boxes. So they want to see enough absorbent material in the box to be able to soak up every drop of water packed if all the bags break open. Calls for a lot more (quite a lot more) newspaper than any shipper actually uses...so you need that sample box to be packed better than real life practice

I would definitely follow the protocol regarding the proper paperwork needed to ship via FedEx. This way, if FedEx ever losses or delays a shipment, you'll actually get reimbursed for the lose. Shipping next day priority is extremely expensive these days, I haven't been able to ship a box for less than $65. The very first priority would be to setup an account so that you qualify for their discounted rates. Obviously thats dependent on how often you ship.
