what size glass....?

Looks like you are leaning toward the glass, so:

1/2" glass all around, no need to temper any of the panels from what I've read.

I think that the sides on top of the bottom allow the silicon to "grip" better and better the sides from blowing out, but I'm not sure of that. I do know that most glass tanks have the side panels in between the front and rear panels, and than all of those sit on top of the base.

Eurobrace should be glass, as sgarron stated, you won't get a bond on acrylic.

I think that with that size tank, a 2" frame would be more than good. You might want to add a certain brace as well though. Thicker won't hurt, and none of this is based on true experience, just observation. Again, the brace should probably sit on top, rather than inside the walls.

I have no clue on that, but I would THINK you could probably go slightly smaller. I would go with more than 1/4" though, if it were me. Maybe 5/16".
 
I have some glass that you could use to build that and probably another one with starfire glass on the viewable pains I will let you have all the glass for $50.

I am not sure how much I have I know it is enough for a 180 gal and some other stuff to but I have it all stacked up I know you would have what you need. Have you ever worked with glass though?
