Shims if you need to level it. Otherwise, no

I would consider attaching a plywood bottom so that you can then seal around the inside edges to contain spills.

I would put "pond liner" inside the stand and wrap it up about 3in on all sides. This gives you some safety for the inevitable water spills around the sump.

Is that a real wood floor or tile that looks like wood?

If it's a real wood floor I'd want some serious water control under and even 2 or 3 feet out in front of the tank. And a liner under the sump, but the Red Sea sump fits so tight you really can't... which is why I got a CADlight system instead.

Seriously, a friend had wood floors and over the years they became a HUGE mess due to small spills and splashes.
