I would stay away from MDF as it swells like a sonofagun with very little water.

I think if you use a 2X6 for the top frame you will be fine without a center support; but you should have one of the experts to confirm.

I would go with 2x8s just to be sure. I think with really good lumber, 2x6s are fine, but I hear good carpenters and other craftsmen are lamenting the quality of lumber nowadays...

I think the point of using the 2x6 was (for a 6 foot tank) really was to avoid a center brace. The 2x4 would work, but would need a brace.
Personally, to go without a center brace, I'd go for a 2x8. Maybe overkill, but a 6 foot tank is borderline 2x6s without a center brace, so I'd size up (to 2x8).

If you place legs at 1' and 5' you will effectively have the same design as on the first page. As such, 2X4s for a 75g will be just fine
