It very much depends on the type of glass. Most aquariums are made of tempered glass which is difficult to impossible to drill and can shatter quite easily. Just Google "drill glass aquarium" for lots of input on this.
i drilled 5 holes in my 180 with a cordless drill and a diamond bit from brs. piece of cake. just build a dam around the hole with plumbers putty then add some water for the bit. dont put too much pressure while drilling...let the bit do the work...also put some duct tape on the inside of the tank where the hole will be. that will catch the plug so it doesnt fall and chip the glass on the other side
If you take your time, use the right bit and plenty of water to keep things cool, then it is fairly simple and the odds of cracking the glass are slim - but not zero! Tempered glass cannot be drilled.
I drilled two holes for 2" bulkheads and one for a 1.5" bulkhead in my 220g. Biggest thing is take your time and let the bit do the cutting and always have water between the bit and glass. They make a guide that suctions to the tank and makes the drilling easier. Also use plumbers putty to make a dam to keep yourself from having a hose or someone else constantly pour water onto the tank
