They do not typically need to be calibrated, I don't think. You use them in every stage of the process - test your tap, test your RO pre DI, test your RODI. That way you know how everything is working.
 
My TDS meter arrived today. I tested a cup of tap water and it was 144ppm. I then tested a bucket of rodi water that I ran last night and it tested 002ppm. Is this acceptable? Do most folks run their filter until it begins to read 000 before running their water change water?

I think 002 is fine. You'll find some TDS when you fill a buckets that you use for mixing salt or doing water changes.

When I used to fill 5 gallon batches I used to run off some of the initial product water. I fill 30 gallon batches now, and don't worry about it.
 
Same here...I make 15 gallons of water at once and don't worry about flushing the RO/DI. If your water changes are just a few gallons, I'd flush it first.

That's all you need it for. To make sure that each section of your RODI is doing what it's supposed to and to figure out what needs to be replaced.
