When I was spawning bettas I kept the male and female separated until she was gravid and ready to go. I've seen males kill females who weren't ready to breed.
the female does have a very large belly. but i don't know what other signs besides darkening and getting stripes or something. but she hasn't darkened or gained any stripes.
